NADPH oxidase controls phagosomal pH and antigen cross-presentation in human dendritic cells Running title: Phagosomal pH regulation in human DC

چکیده
The phagocyte NADPH oxidase (NOX2) is critical for the bactericidal activity of phagocytic cells and plays a major role in innate immunity. We showed recently that NOX2 activity in mouse dendritic cells (DC) prevents acidification of phagosomes, promoting antigen crosspresentation. In order to investigate the role of NOX2 in the regulation of the phagosomal pH in human DC, we analyzed the production of reactive oxygen species (ROS) and the phagosomal/endosomal pH in monocyte-derived DC and macrophages (MØ) from healthy donors or chronic granulomatous disease (CGD) patients. As expected, we found that human MØ acidify their phagosomes more efficiently than human DC. Accordingly, the expression of the vacuolar proton ATPase (V-H+-ATPase) was higher in MØ than in DC. Phagosomal ROS production, however, was also higher in MØ than in DC, due to higher levels of gp91phox expression and recruitment to phagosomes. In contrast, in the absence of active NOX2, the phagosomal and endosomal pH decreased. Both in the presence of a NOX2 inhibitor and in CGD patients-derived DC, the cross-presentation of two model tumor antigens was impaired. We conclude that NOX2 activity participates in the regulation of the phagosomal and endosomal pH in human DC, and is required for efficient antigen cross-presentation. متن کاملPhagosomal proteolysis in dendritic cells is modulated by NADPH oxidase in a pH-independent manner.
The level of proteolysis within phagosomes of dendritic cells (DCs) is thought to be tightly regulated, as it directly impacts the cell's efficiency to process antigen. Activity of the antimicrobial effector NADPH oxidase (NOX2) has been shown to reduce levels of proteolysis within phagosomes of both macrophages and DCs. However, the proposed mechanisms underlying these observations in these tw...
متن کاملNADPH oxidase activity controls phagosomal proteolysis in macrophages through modulation of the lumenal redox environment of phagosomes.
The phagosomal lumen in macrophages is the site of numerous interacting chemistries that mediate microbial killing, macromolecular degradation, and antigen processing. Using a non-hypothesis-based screen to explore the interconnectivity of phagosomal functions, we found that NADPH oxidase (NOX2) negatively regulates levels of proteolysis within the maturing phagosome of macrophages. Unlike the ...
متن کاملCLIC1 regulates dendritic cell antigen processing and presentation by modulating phagosome acidification and proteolysis
Intracellular chloride channel protein 1 (CLIC1) participates in inflammatory processes by regulating macrophage phagosomal functions such as pH and proteolysis. Here, we sought to determine if CLIC1 can regulate adaptive immunity by actions on dendritic cells (DCs), the key professional antigen presenting cells. To do this, we first generated bone marrow-derived DCs (BMDCs) from germline CLIC1...
متن کاملIntracellular Regulation of Cross-Presentation during Dendritic Cell Maturation
We have investigated the effect of different maturation stimuli on the ability of mature dendritic cells (DCs) to cross-present newly acquired particulate antigens. Cross-presentation was impaired in DCs matured by treatment with TNF-α, CpG and LPS, but was less affected upon CD40L-induced maturation. The difference could not be explained by decreased antigen uptake or translocation into the cy...
